摘要
多抗甲素是一种新型的免疫增强剂,临床应用证明多抗甲素对再生障碍性贫血疗效较好。为了探讨多抗甲素治疗再障的机理,我们曾用小鼠照射后形成脾集落的方法,对造血干细胞CFU-S作了测定,发现多抗甲素有刺激造血的作用。在此基础上,我们应用电子显微镜技术,对CFU-S脾集落细胞进行了电镜观察。本文首次报告了CFU-S脾集落细胞的超微结构特征。发现多抗甲素对脾集落细胞的超微结构有明显影响,能促进脾集落细胞增殖分化,脾集落细胞显示出代谢旺盛、功能活跃、DNA及蛋白质合成增加等表现。本文根据免疫缺陷学说对再障发病机理作了探讨,从超微结构水平讨论了多抗甲素对造血干细胞的影响。
Electron microscopic study on the colonies cells of spleen were presented.
Polyactin A is a new immunological stimulant which was developped in China for the first time. It is a polysaccharide extracted from cultured of α-hemolytic streptococcus No. 33 in the mouth. Clinical abservation suggested that the drug had significant inhibiting effects on some tumors and could increase the number of leucocytes, and stimulate immunity of the body organism. Polyactin A had also a good effect on aplastic anemia 'and it was being used as a new drug in the treatment of aplastic anemia, but its mechanism wasn't clear. In order to investigate this problem, electrn microscopic observation on the colonies cells of spleen (CFU-s) were studied after effects of Polyactin A on hemopoietic stem cells of marrow were studied. The results showed that Polyactin A could make marked effects on ultrastructures of CFU-S.
Effects of Polyactin A on hemopoietic stem cells were disccused at ultrastructural level in the study and mechanisms of aplastic anemia were also evaluated.
