摘要
目的探讨单纯形优化方法在腺苷脱氨酶最佳反应条件选择中的作用。方法运用单纯形优化法对腺苷脱氨酶最佳反应条件进行筛选,参考相关文献确定初始单纯形,根据各种组合条件下得到的测定结果进行单纯形的移动与终止,从而得到最佳反应条件。结果 8种影响因素组成的单纯形进行了24次移动得到33种反应条件的组合。腺苷脱氨酶样本在第28种组合条件下得到的测定结果最高,该反应条件即为最优条件,该条件下测定活性比目前推荐的测定方法提高近1倍。结论单纯形法是寻找临床酶学测定最优反应条件的一种简单有效的方法。
Objective To explore the key role of the simplex optimization method for optimized reaction condition selection of adenosine deaminase(ADA). Methods The simplex optimization method was used to select the optimized reaction condition for ADA.The original simplex was fixed by pertinent literature.The simplex was moved or stopped according to the results obtained under the all kinds of combination,after that we could get the optimal reaction conditions. Results The results were found in 33 tests after 24 movements of the simplex which consisted of 8 factors.We found the optimum formulation was combination 28,in which we could get the maximum catalytic activity concentration of ADA. Conclusions The simplex method seems to be one of the simple and effective techniques for reaction condition selection of clinical enzymes.
