固相萃取—HPLC-PDA法测定猪肉中的盐酸克伦特罗的研究

摘要 对固相萃取——HPLC-PDA法测定猪肉中的盐酸克伦特罗的前处理方法和色谱条件进行了优化。超声提取时间30min,固相萃取柱净化过柱速度为2d/s。色谱条件为V甲醇∶V0.01mol/L磷酸二氢钠溶液=35∶65,固定相ODS-C18,高效液相色谱柱4.6μm×150mm,流速1mL/min,检测波长243nm。优化试验条件后盐酸克伦特罗残留量含量回收率得到较大改善,该方法检测线性良好,操作简单、快速,色谱峰形理想,结果准确可靠,重现性佳。 In this paper, solid-phase extraction--RP-HPLC was applied for the detection of clenbuterol residue in pork and both the pre-treatment method and chromatographic conditions were optimized. For the pre-treatment of the meat samples, the best ultrasonic extraction time and elution speed of the extracts on solid phase were 30 minutes and 2d/s, respectively. Then the purified extracts was analyzed by RP-HPLC on a ODS C-18 column ( 4.6 μm × 150 mm)using 0.01mol/L of solium dihydrogen phosphate 1mL/min and a UV detector at 243nm. This method residual recovery, and good reproducibility. solution-methanol(65 : 35)as mobile phase at a flow rate of for clenbuterol residue detection in pork had high accuracy,residual recovery, and good reproducibility.
