摘要
目的研究西妥昔单抗(C225)对不同照射模式人鼻咽低分化鳞癌细胞(CNE-2)裸鼠移植瘤细胞凋亡相关基因Bax、Bcl-2表达的影响。方法成功构建48只CNE-2裸鼠移植瘤模型,待成瘤后随机分为空白对照组、急速照射组、模拟IMRT组、C225组、急速照射+C225组、模拟IMRT+C225组,运用RT-PCR法检测Bax、Bcl-2的转录水平。结果 Bax转录水平:空白对照组、急速照射组、模拟IMRT组、C225组、急速+C225组和模拟IM-RT+C225组转录水平分别为0.452、0.662、0.558、0.542、0.621、0.555,放疗后Bax表达上调,急速照射组表达高于模拟IMRT组(P=0.005);使用C225后Bax表达上调(P=0.015);急速与急速+C225组、模拟IMRT组与模拟IMRT+C225组差异均无统计学意义(P>0.05)。Bcl-2转录水平:空白对照组、急速照射组、模拟IMRT组、C225组、急速+C225组和模拟IMRT+C225组转录水平分别为0.629、0.685、0.654、0.625、0.658、0.648,各组差异均无统计学意义(P>0.05)。结论模拟IMRT照射时间延长,影响Bax的表达;C225可能具有一定促凋亡作用;放疗联合C225同单独使用放疗相比Bax表达无明显差异;照射及应用C225后Bcl-2表达的相对稳定,不受照射方式及C225的影响。
Objective To explore the antitumor effect of different radiation pattern and investigate it of C225 combining different radiation pattern on human nasopharyngeal carcinoma CNE-2 xenografts in nude mice.and detect the expressions of Cell apoptosis genes of Bax and Bcl-2.Methods CNE-2 cells were injected into the 48 nude mice,the mice were divided into 6 groups randomly: control group,acute irradiation model group,IMRT group,C225 group,acute irradiation model+C225 group,IMRT+C225 group.Each group had 8 nude mice.RT-PCR was used to detect mRNA expressions of cell apoptosis genes of Bax and Bcl-2.Results Expresssion of Bax: The expression of those groups were 0.452,0.662,0.558,0.542,0.621 and 0.555 respectively;the expression of acute irradiation model group was higher than IMRT group(P=0.005);Only use C225 can up the expression of Bax.But there was no significant difference between acute irradiation model group and acute irradiation model+C225 group,IMRT group and IMRT+C225 group.Expresssion of Bcl-2.The expression of those group were 0.629,0.685,0.654,0.625,0.658 and 0.648 respectively.There was no significant difference between every two groups(P0.05).Conclusion IMRT model prolongs fraction delivery time,and affects the expression of Bax.Only use C225 have some effect of inducing apotosis,but combine radiotheraphy have no change than only radiotherapy.Wether radiotherapy or combine C225 will not change the expression of Bcl-2.
