摘要
目的建立HPLC法测定美洲大蠊药材中氨基酸含量的方法。方法用盐酸水解、邻苯二甲醛和β-巯基乙醇柱前衍生,0.50%磷酸二氢钠缓冲液(pH7.8)-甲醇-乙腈为流动相,柱温:35℃,检测波长:338 nm,梯度洗脱后测定氨基酸的含量。结果各氨基酸衍生物在40 min内均能得到良好的分离,r值为0.999～1.000 0(n=6),平均回收率为92.34%～103.9%,RSD为1.18%～4.76%。结论该方法简便、准确、重复性好,可作为美洲大蠊中氨基酸的含量测定。
Objective To establish an HPLC method for determining amino acids in Periplaneta americana. Methods After being hydrolyzed with HCl and derivated in precolumn with o-phthalaldehyde and β-mercaptoethanol,the amino acids were eluted by gradient elution and were detected with an UV detector（detection wavelength：338nm）.The mobile phase consisted of 0.55% NaH2PO4（pH 7.80）-methanol-acetonitrile,column temperature was set at 35 ℃. Results The derivatives of amino acids with o-phthalaldehyde could be separated with a stable base line within 40 min.The calibration curves for amino acids had good linearity（r=0.999 0~1.000 0,n=6）,the average recoveries were 92.34%~103.9%,with RSDs of 1.18 %~4.76%. Conclusion The method is simple,accurate and reproducible for the determination of amino acids in Periplaneta americana.
