康莱特对HepG2细胞凋亡及其Caspase-3和Survivin表达影响 被引量:4

EFFECTS OF KANGLAITE INJECTION ON APOPTOSIS OF HEPG2 CELLS AND EXPRESSIONS OF CASPASE-3 AND SURVIVIN

摘要 目的观察康莱特注射液对HepG2肝癌细胞凋亡及半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)和存活素(Survivin)表达的影响,并初步探讨其作用机制。方法分别采用体积分数0.005、0.010、0.015、0.020的康莱特体外培养HepG2细胞。采用MTT法检测HepG2细胞抑制率,流式细胞仪检测HepG2凋亡情况以及Caspase-3和Survivin表达的变化。结果康莱特能明显抑制HepG2肝癌细胞生长,且呈时间和浓度依赖性。康莱特组中Caspase-3、Survivin蛋白的表达与对照组比较,差异有显著性(P<0.01)。结论康莱特可抑制HepG2肝癌细胞增殖,并可通过提高Caspase-3蛋白表达和降低Survivin蛋白表达诱导HepG2肝癌细胞凋亡。 Objective To investigate the effects of Kanglaite injection(KLT) on apoptosis of hepatoma carcinoma cell HepG2(HepG2),expressions of Caspase-3 and Survivin,and discuss their mechanisms.Methods HepG2 cells were cultured in vitro with different concentrations of KLT(0.005,0.010,0.015,and 0.020),cell inhibition rate was detected by MTT method,and apoptosis of HepG2,expressions of Caspase-3 and Survivin were determined by using flow cytometry.Results KLT obviously inhibited the growth of HepG2,manifesting time-and concentration-dependent.In KLT group,the differences of expressions of Caspase-3 and Survivin were significant versus the control group(P0.01).Conclusion KLT can inhibit HepG2 cell multiplication,and induce apoptosis of HepG2 by increasing expression Caspase-3 and decreasing the expression of Survivin.
