摘要
目的研究在非小细胞肺癌(non-small cell lung cancer,NSCLC)中,Axin(Axis inhibitor)与细胞凋亡的关系以及X射线能否上调NSCLC组织中Axin表达,并促进NSCLC细胞凋亡。方法用免疫组织化学和TUNEL(TdT-mediated dUTP-X射nick end labeling)法检测95例NSCLC组织内Axin的表达及凋亡情况。将新鲜的NSCLC组织及培养的细胞系(A549和BE1)用X射线照射后,用免疫印迹和RT-PCR方法检测组织中Axin、凋亡蛋白caspase-3表达,并用流式细胞仪检测细胞凋亡情况。结果在hNSCLC组织内,可见少量凋亡细胞和Axin阳性细胞,凋亡与Axin的表达呈正相关。X射线照射后,部分病例的hNSCLC组织内Axin表达明显上调并促进A549和BE1细胞系凋亡。结论 X射线能诱导NSCLC组织Axin表达上调并促进A549和BE1细胞凋亡。
Objective To study the correlation of Axin(Axis inhibition) with apoptosis and the effect of X-ray on Axin expression and apoptosis in non-small cell lung cancer(NSCLC).Methods Immunohistochemistry and TUNEL(TdT-mediated dUTP-X nick end labeling) were used to detect the expression of Axin and apoptosis in NSCLC tissues.Western blotting and RT-PCR were used to detect Axin and caspase-3 expression in NSCLC tissues,and flow cytometor was used to detect apoptosis in A549 and BE1 cell line.Results The weak positive Axin and apoptosis cells were found with positve correlation for the expression level of Axin and apoptosis in NSCLC tissues.The expression level of Axin was upregulated in apart of NSCLC tissues,and apoptosis was enhangced more obviously in Axin high expression A549 and BE1 cell line.Conclutions X-ray upregulates Axin expression and induces apoptosis more obviously in Axin high expression A549 and BE1 cell line.
