摘要
目的:研究针对TGF-βⅡ型受体的RNA干扰质粒对α-SMA表达的抑制作用,探讨RNA干扰TGF-βRII的方法对肾间质纤维化的抑制作用。方法:单侧结扎输尿管方法制备肾间质纤维化小鼠动物模型。分别经输尿管逆行注射a:RNA干扰质粒;b:错配对照质粒;c:空载体;d:生理盐水;e:正常对照。通过western-blot及免疫组织化学方法检测第3、5、10天后肾组织内TGF-βRⅡ、α-SMA,观察其对肾间质纤维化的抑制作用。结果:针对TGF-βRII的RNA干扰质粒,明显抑制肾组织内TGF-βRⅡ、α-SMA表达;几组对照未见相应作用。结论:针对TGF-βRⅡ的RNA干扰质粒,能够抑制肾间质纤维化的发生、发展,可能成为延缓肾功能减退的有效方法。
Objective: To investigate the inhibitory effect of RNA interference against TGF-β Ⅱreceptor(TGF-β R Ⅱ) on α-SMA expression and renal fibrosis.Methods:The Unilateral ureteral obstruction(UUO) model which was made by using an established procedure was performed for the injected mice to evaluate the anti-fibrotic effect.The formulation includes: pSUPER-TGF-β RII(100μg/mouse),mismatch pSUPER-TGF-β RII(100μg/mouse),empty pSUPER(100μg/mouse),saline,nothing.The inhibition effect of the siRNA about TGF-β RⅡ on renal fibrosis were detected.TGF-β RII and α-SMA were detected by western-blot and immunol histochemistry at 3d,5d and 10d after the injection.Results:The injection of plasmid DNA of pSUPER-TGF-β RⅡ decreased significantly the level of TGF-β RII,α-smooth muscle actin(a-SMA) and collagen type I over-expression incontrast to free plasmid DNA injection.Conclusion: Retrograde injection of TGF-β R siRNA expression vector plasmid DNA is available to suppress progression of renal interstitial fibrosis.
