摘要
目的研究轮状病毒(Rotavirus,RV)P[8]G1和P[2]G3株灭活疫苗的免疫原性及不同血清型间的交叉识别作用。方法采用Vero细胞转瓶培养RV,经纯化灭活、佐剂吸附等工艺,制备RV P[8]G1和P[2]G3株灭活疫苗,分别经肌内免疫Wistar大鼠和猕猴,采用ELISA法和微量病毒中和试验检测大鼠和猕猴血清中RV特异性抗体效价和中和抗体效价,流式细胞术检测猕猴血液中CD4+和CD8+T细胞的数量。结果 P[8]G1和P[2]G3株疫苗样品的RV抗原含量分别为10 240和5 120 EU/ml;免疫2次后,P[8]G1和P[2]G3株疫苗诱导大鼠产生的血清IgG抗体GMT分别为(43 407.71±1 654.51)和(41 520.61±2 047.61),诱导猕猴产生的血清IgG抗体GMT分别为(13 654.45±3 125.27)和(10 350.68±997.83);单一血清型疫苗免疫后产生的中和抗体具有型间交叉识别作用;2株疫苗免疫猕猴后均能刺激其CD4+T淋巴细胞增殖,CD8+T淋巴细胞无明显变化。结论 2株RV灭活疫苗均具有良好的免疫原性及型间交叉识别作用。
Objective To investigate the immunogenicity and cross protective effects of inactivated rotavirus(RV) vaccines prepared with P[8]G1 and P[2]G3 strains.Methods RV P[8]G1 and P[2]G3 strains were cultured in Vero cells in rolling bottle,then purified,inactivated and adsorbed with adjuvant to prepare inactivated vaccine respectively.Wistar rats and rhesus macaques were immunized with the prepared vaccines separately,of which the RV specific antibody and neutralizing antibody titers in sera were determined by ELISA and microneutralization assay respectively.The CD4+ and CD8+ T lymphocytes in blood of rhesus macaques were counted by flow cytometry.Results The RV antigen contents in vaccines prepared with P[8]G1 and P[2]G3 strains were 10 240 and 5120 EU / ml respectively.The serum GMTs of IgG induced by two doses of vaccines prepared with P[8]G1 and P[2]G3 strains were(43 407.71 ± 1 654.51) and(41 520.61 ± 2 047.61) in rats while(13 654.45 ± 3 125.27) and(10 350.68 ± 997.83) in rhesus macaques,respectively.The neutralizing antibody induced by vaccine of a single serotype showed cross protective effect against various serotypes.Both the vaccines prepared with two strains stimulated the proliferation of CD4+ T lymphocytes in rhesus macaques,while showed no significant effect on proliferation of CD8+ T lymphocytes.Conclusion Both the vaccines prepared with P[8]G1 and P[2]G3 strains showed good immunogenicity and cross protective effect against various serotypes of RV.
