油菜蜂花粉水溶性蛋白的分离及其抗氧化活性 被引量:2

Study on Isolation and Antioxidant Activity Analysis of Water-soluble Protein from Rape Bee Pollen

摘要 采用饱和硫酸铵法对油菜蜂花粉中的水溶性蛋白进行逐级分离提取,发现20%~40%浓度的硫酸铵对蜂花粉水溶性蛋白的分离效果最好,得率和回收率分别为711.54μg/g和29.91%;同时,对不同硫酸铵浓度(0%~20%、20%~40%、40%~60%、60%~80%和80%~100%)得到的水溶性蛋白的体外抗氧化活性进行测定,发现20%~40%浓度硫酸铵盐析得到的蛋白对DPPH和·OH自由基的清除能力最强;相同浓度下,各级水溶性蛋白对·OH自由基的清除能力均强于DPPH自由基。结合SDS-PAGE图谱分析表明,分子质量约为39.0 ku和约为58.0 ku的2种蛋白有可能在油菜蜂花粉水溶性蛋白的抗氧化反应中发挥着重要的作用。 Rape bee pollen water-soluble proteins (RBPWPs) were isolated by the process of the ammonium sul- fate salting-out. The results showed that isolation with 20% - 40 % concentration of ammonium sulfate is the best, the yield and recovery were 711.54 Ixg/g and 29.91% , respectively. Also, antioxidative activities of RBPWPs with different concentration of ammonium sulfate precipitation (0% -20 % , 20% -40 % , 40% -60 % , 60% - 80 % and 80% - 100 % ) were determined in vitro. DPPH and · OH radical scavenging assay showed that RBPWPs with 20% - 40 % concentration of ammonium sulfate had the highest activity. Additionally, all RBPWPs scavenged ·OH radical activities were higher than DPPH. Combination with SDS-PAGE result, it indicated that RBPWPs with -39. 0 ku and ·58.0 ku molecular weight may play an important role in antioxidation reactions.
