响应面法优化巨大芽孢杆菌中的聚-β-羟基丁酸(PHB)提取工艺 被引量:1

Optimal Extraction Conditions of Poly-β-hydroxybutyrate(PHB) in Bacillus megaterium by Response Surface Methodology

摘要 为获得聚-β-羟基丁酸(PHB)提取的最佳工艺,以氯仿浓度,提取时间,和提取温度为影响因子,在单因素实验结果的基础上,应用Box-Benhnken中心组合方法进行三因素三水平的实验设计,以PHB提取量为响应值,运用响应面法(RSM)对提取条件进行进一步的优化。结果表明:在氯仿浓度69.7%,提取温度59.7℃,提取时间3.0 h的条件下,PHB提取量达到最大值4.430 g/L。 To obtain the optimal extraction conditions of Poly-β-hydroxybutyrate (PHB) in Bacillus megaterium, the effect of chloroform concentration, extraction time and temperature on PHB extraction were studied. On the basis of single factor experiment, Box-Benhnken central composite method was designed for the three factors. In response to PHB extrac- tion value, the extraction conditions were further optimized by response surface method (RSM). The results showed that when chloroform concentration was 69.7% ,extraction temperature was 59.7 ~C ,extraction time was 3.0 h,the produc- tivity of PHB was 4.437 g/L, reached its maximum.
