胃癌组织中CXCL12和CXCR4的表达及意义 被引量:7

摘要 目的观察趋化因子CXCL12及其特异性受体CXCR4在人胃癌组织中的表达,探讨其与临床病理参数、预后的关系。方法选择120例胃癌标本,应用免疫组化SP法检测CXCL12和CXCR4在人胃癌组织中的表达,分析CXCL12和CXCR4的表达与患者临床病理参数、术后生存率之间的关系。结果胃癌组织及正常胃黏膜组织中均可检测到CXCL12、CXCR4的表达,但胃癌组织中的表达水平均明显高于正常胃黏膜组织,表达差异有显著性(P<0.05)。CXCL12阳性与CXCR4阳性呈正相关(r=0.276,P<0.05)。胃癌CXCL12和CXCR4的表达水平与肿瘤细胞淋巴结转移及分化程度密切相关(P<0.05),与患者的年龄、性别、肿瘤的大小、浸润深度及远处转移等无关(P>0.05)。CXCL12和CXCR4阳性表达的患者其五年生存率明显低于其阴性表达的患者。结论胃癌中CXCL12和CXCR4的高表达与胃癌的生物学行为及预后密切相关,检测其表达对预测胃癌的转移及判断预后有一定价值。 Purpose To investigate the expressions of chemokine CXCL12 and its receptor CXCR4 protein in gastric cancer and the relationship between clinicopathologic parameters and the prognosis.Methods Immuohistochemistry was used to detect the expressions of CXCL12 and CXCR4 in 120 samples of human gastric cancer.The correlation between the expression of CXCL12,CXCR4 and clinicopathologic parameters of the patients with gastric cancer was analyzed respectively.To analyzed the value of CXCL12,CXCR4 and combined assay of the two proteins in predicting the prognosis of patients.Results CXCL12 and CXCR4 were detected in both gastric carcinoma tissues and normal tissues,but the expression level of CXCL12 and CXCR4 were found up-regulated in gastric carcinoma tissues as compared with normal tissues.The expression of CXCL12 and CXCR4 are significantly different(P0.05).The positive expression of CXCL12 was consistent with the positive expression of CXCR4(r=0.276,P0.05),and the expression levels of CXCL12 and CXCR4 were closely correlated with lymph node metastasis and differentiation(P0.05),but not with age,sex,tumor size,depth of invasion,distant metastasis(P0.05).The five-year survival rate of patients whose CXCL12 and CXCR4 positive expression was significantly lower than the patients with negative expression.Conclusion Overexpression of CXCL12 and CXCR4 is correlated with the biologic behavior and prognosis of gastric carcinoma,determination of CXCL12 and CXCR4 is very helpful in predicting of metastasis and evaluating the prognosis of gastric carcinoma.
