摘要 目的通过扩充基质辅助激光解吸电离飞行时间质谱(matrixassisted laser desorption ionization-time of flight mass spectrometry,MALDI-TOF-MS)的数据库研究宋内志贺菌的鉴定。方法用提取法进行宋内志贺菌质谱图的建库,建库后分别使用直接涂抹法和提取法对50株宋内志贺菌野生株进行鉴定。结果提取法的鉴定符合率为100%,而直接涂抹法的鉴定符合率为60%。结论 MALDI-TOF-MS通过数据库的扩充和流程的优化可以鉴定宋内志贺菌到血清学水平,它将会成为临床微生物实验室的诊断和鉴别工具。 Objective To investigate the identification of Shigella sormei through expanding the database of matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF-MS). Methods Extraction method was used to create the database of mass spectrum map of Shigella sonnei, and then smear method and extraction method were used for the identification of 50 wild strains of ShigeHa sonnei. Results Identification coincidence rates were 100% using extraction method, and 60% using smear method. Conclusions MALDI-TOF-MS can be used for serological screening for the identification of Shigella sonnei by exlmading the database and optimizing the procedure, and it will become the tool for the diagnosis and differentiation in clinical microorganism laboratory.
