摘要
目的探讨褪黑素(MT)对脂多糖(LPS)诱导的内毒素血症大鼠循环血和血管组织中肿瘤坏死因子-α(TNF-α)表达的影响。方法雄性SD大鼠,分组如下:(1)溶剂对照组;(2)LPS组;(3)LPS+MT组;(4)MT组。应用酶联免疫吸附法检测血清TNF-α含量;应用RT-PCR和免疫组织化学法检测TNF-α在血管组织的表达。结果与对照组相比,LPS组大鼠血清TNF-α含量、血管TNF-α mRNA水平均增高(P<0.01),TNF-α蛋白在血管内皮和平滑肌细胞阳性表达增多;而给予LPS前后应用MT干预治疗,可显著降低血清TNF-α含量、血管TNF-α mRNA生成,与同时间点LPS组比较均差异有统计学意义(P<0.01)。结论 MT降低内毒素血症大鼠TNF-α生成和表达。
Objective To investigate the influence of melatonin(MT) on the expression of tumor necrosis factor alpha (TNF-ot) in peripheral blood and vascular tissue of rats with endotoxemia induced by lipopolysaccharides(LPS). Methods The male SD rats were divided into four groups:solvent control group;LPS group;LPS + MT group;MT group(5 mg/ml,i, p. ). The content of TNF-α in serum was detected by enzyme linked immunosorbent assay(ELISA). The expression of TNF-cxmRNA in vascular tissue was detected by RT-PCR and immunohsitochemistry. Results As compared with those in control group, the levels of TNF-ct in serum and the expression of TNF-α mRNA in vascular tissue were significantly increased in LPS group ( P 〈 0.01 ), and the expression of TNF-α protein in vascular endothelium and smooth muscle cells was increased, however, the MT intervention therapy before and after administrating LPS could obviously reduce the content of TNF-α in serum, and could decrease the production of TNF-α mRNA in blood vessel in LPS + MT group, as compared with those in LPS group at the same time point( P 〈 0.01 ). Conclusion MT can inhibit the generation and expression of TNF-α in rats with endotoxemia.
