葛根素改善高胆固醇血症兔缺氧及其机制的研究

摘要 目的探讨葛根素(Pu)对高胆固醇血症(HC)所致兔缺氧的改善及其作用机制。方法新西兰兔分3组:对照组、HC组和葛根素治疗(HP)组,分别喂饲普通饲料、高胆固醇饲料和高胆固醇加Pu饲料。3周后检测血清总胆固醇(TC)含量、静注乙酰胆碱(ACh)前后动静脉血血气分析及血压、肾组织血管通透性、胸主动脉环ACh介导的舒张反应、脑匀浆一氧化氮合酶(NOS)和超氧化物歧化酶(SOD)的活力及一氧化氮(NO)和丙二醛(MDA)的含量。结果Pu可明显改善HC所致的血氧饱和度等血气分析指标及ACh降压效应的异常;Pu能降低血清TC、脑组织MDA含量、NOS活力以及肾组织血管通透性,使脑组织NO生成增多、SOD的活力增大;2组ACh介导的胸主动脉环舒张功能均较对照组降低但未达统计学显著性。结论Pu可能通过降血脂、改善血管内皮依赖性舒张功能、降低血管通透性等作用,改善机体缺氧,从而抑制其对NOS活性的上调,降低机体氧化应激水平及其造成的组织损害。
