摘要
目的:探讨重组腺病毒Rb94(Ad-Rb94)基因转染对宫颈癌细胞辐射敏感性的影响。方法:将Ad-Rb94基因转染宫颈癌细胞株Hela,进行137Csγ射线照射,数字随机表法分为空白对照组、Ad-LacZ组、Ad-Rb94组、照射组和Ad-Rb94联合照射组。用RT-PCR法和免疫荧光法检测Rb-94基因mRNA和蛋白水平的表达,以MTT法观察细胞的生长曲线和单细胞凝胶电泳(SCGE)实验观察细胞DNA的损伤。结果:RT-PCR法和免疫荧光法检测到在Hela细胞有Rb-94基因的表达,Ad-Rb94组、照射组和Ad-Rb94联合照射组Hela细胞的生长均受到抑制,其中联合照射组的抑制效应最强(P<0.05)。照射组和联合照射组细胞照射后出现DNA损伤,联合组损伤程度强于照射组,差异有统计学意义(P<0.05)。结论:Ad-Rb94基因联合照射能明显增强人宫颈癌细胞的辐射敏感性。
OBJECTIVE: To study the radiation sensitivity effects of Ad-Rb94 gene combined with ionizing radiation on Hela cells.METHODS: Hela cells were transfected with Ad-Rb94 gene and irradiated by 137Cs γ-ray.The cohort randomly was divided into blank control,Ad-LacZ,Ad-Rb94,radiation and Ad-Rb94 combined with radiation groups.The mRNA and protein expression of Ad-Rb94 gene were detected by RT-PCR and immunofluorescence method,the cell growth curve was observed by MTT assay and the DNA damage was analyzed by SCGE technique.RESULTS: The result of RT-PCR and immunofluorescence showed that the Rb-94 gene was presented in Hela cells.The growth of Hela cells transfected with Ad-Rb94,radiation and Ad-Rb94 combined with radiation group was all inhibited.The group of Ad-Rb94 combined with radiation resulted in greatest inhibition of cells growth(P0.05).DNA damage in Ad-Rb94 combined with radiation group was significantly higher than that in radiation group(P0.05).CONCLUSION: Ad-Rb94 combined with radiation can apparently increase the radiation sensitivity effects on Hela cells.
