摘要
目的:采用RP-HPLC法测定排风藤茎叶果中番茄碱的含量.方法:建立RP-HPLC法测定排风藤茎、叶、果中番茄碱含量的方法.色谱柱为YMC-Pack ODS-AA(150mm×4.6mm,5μm),流动相为乙腈-水(90∶10,v.v-1),流速为1mL.min-1,柱温为30℃,紫外检测波长为203nm.线性范围:0.10~0.60mg.mL-1(相关系数r=0.999 7).结果:测得排风藤茎叶果中番茄碱的含量分别为1.104mg.g-1、0.494mg.g-1、1.030mg.g-1;RSD分别为2.60%、2.40%、2.80%;平均回收率为97.6%,RSD为2.93%(n=9).结论:该方法快速、简便、准确度高,可为开发利用三峡库区民间中草药排风藤提供研究基础.
The contents of tomatine in stems,leaves and fruits of Solanum cathayanu were determined by RP-HPLC,using external reference method.RP-HPLC determination was established.The analyte was separated on a YMC-Pack ODS-AA column(150 mm × 4.6 mm,5 μm) at 30 ℃ with mobile phase contained acetonitrile-water(90 : 10,v·v-1) at a flow rate of 1 mL·min-1 followed by UV detection at 203 nm.The calibration curve was well linear over the concentration range from 0.10 to 0.60 mg·mL-1(r=0.999 7);and the limit of detection was 0.01 mg·mL-1.The contents of tomatine in stems,leaves and fruits of Solanum cathayanu are 1.104,0.494,and 1.030 mg·g-1,respectively.The average recovery is 97.6% with a RSD of 2.93%(n=9).This method is fast,simple and accurate,and can provide the theoretical basis for further exploration of Solanum cathayanu.
