IGF-Ⅰ明胶微球对骨髓基质干细胞增殖作用的实验性研究 被引量:1

摘要 目的制备胰岛素样生长因子-Ⅰ明胶微球(insulin-like growth factor-Ⅰ gelatin microspheres,IGF-I-GMs),观察其一般性质、体外释药特性及其对兔骨髓基质干细胞(bone marrow stromal cells,BMSCs)增殖的长期效应。方法用乳化交联法制备IGF-Ⅰ-GMs,观察其一般性质;ELISA法测定IGF-Ⅰ的含量,计算微球包封率和载药率及体外释药特性。体外培养兔BMSCs,四唑盐比色法(methyl thiazolyltetrazolium,MTT)检测IGF-Ⅰ和IGF-Ⅰ-GMs梯度含量分别对BMSCs增殖的长期效应。结果①所制备的微球表面光滑圆整,球体均匀度好,平均粒径为(19.51±3.53)μm,冻干粉剂为黄白色粉末状,再分散性良好,微球载药量和包封率分别为890ng/g和89.0%,体外7d内药物缓释87%;②IGF-Ⅰ和IGF-Ⅰ-GMs均明显促进BMSCs增殖,IGF-Ⅰ-GMs较单独应用IGF-Ⅰ的效果更加显著(P<0.05),呈浓度及时间依赖性。结论 IGF-Ⅰ-GMs冻干粉剂制备良好;IGF-Ⅰ-GMs在7d内对IGF-Ⅰ有良好的缓释作用;BMSCs形态和活性良好可以用于实验研究;IGF-Ⅰ-GMs与BMSCs有良好的生物相容性,且促进BMSCs增殖效果明显优于单独使用IGF-Ⅰ。
