摘要 目的研究转录因子GCM1和GATA-3对Syncytin 1表达的调控作用。方法应用实时RT-PCR检测绒癌细胞JEG-3和人星形胶质细胞瘤CCF-STTG1细胞Syncytin 1及其转录因子mRNA水平,并采用RNA干扰技术探讨GCM1和GATA-3对Syncytin 1转录的调控。结果 Syncytin 1、GCM1和GATA-3的mRNA水平在JEG-3与CCF-STTG1细胞间差异有统计学意义(均P<0.01);流感病毒A感染使CCF-STTG1细胞Syncytin 1、GCM1和GATA-3转录显著增高(均P<0.001);GCM1 siRNA或GATA-3siRNA使JEG-3细胞Syncytin 1的表达显著降低(P<0.05),而CCF-STTG1细胞没有显著改变(P>0.05)。结论 GCM1和GATA-3调控Syncytin 1在绒癌细胞JEG-3中的特异表达,但不足以调控病毒感染诱导的人星形胶质细胞瘤细胞CCF-STTG1的Syncytin 1异常转录。 [Objective] To investigate the roles of transcriptional factors GCM1 and GATA-3 on the cellular specific expression of Syncytin 1. [Methods] The mRNA levels of Syncytin 1 and its transcriptional factors were measured with real time RT-PCR. Furthermore, the possible modulation of GCM1 and GATA-3 on the transcription of Syncytin 1 was explored with RNA interference. [Results] The transcriptional levels of Syncytin 1, GCM1, and GATA-3 were significantly higher in JEG3 cells as compared to CCF-STrG1 cells (all P〈0.01). The mRNA levels of Syncytin 1, GCM1 and GATA-3 in CCF-STFG1 cells elevated significantly with the influenza A infection (all P〈0.001). With GCM1 or GATA-3 RNA interference, the mRNA level of Syncytin 1 in JEG-3 cells reduced significantly (P〈0.05), however, no such effects were observed in CCF- STTG1 cells. [Conclusion] This study suggests that transcriptional factors GCM1 and GATA-3 may regulate the cellular specific transcription of Syncytin 1 in JEG-3 cells, but are not enough for the aberrant expression of Syncytin 1 induced by viral infection in CCF-STrG1 cells.
