摘要
采用紫外化学复合诱变法对纳豆菌原生质体进行诱变,以期获得高抑菌活性的优良菌株.结果表明,在溶菌酶质量浓度为0.3mg/mL、酶解时间为40min、酶解温度为35℃、酶解pH为7.0下原生质体的形成率和再生率达到最高.通过紫外化学复合诱变,得到一株高抑菌活性菌株.与原菌株比较,其对金黄色葡萄球菌的抗菌作用提高了19.7%,对大肠杆菌的抗菌作用提高了19.5%.连续传代培养10代,抗菌活性稳定.
To obtain excellent strain with a high antibacterial activity,the composite mutation of Bacillus natto protoplast was studied by UV ray and 5-fluorouracil.The results showed that under the condition of 0.3 mg/ml enzymolysis concentration,40 min enzymolysis time,35 ℃ enzymolysis temperature and pH 7.0 enzymolysis,the pretoplast formation and regeneration rates reached its highest.By compound UV and chemical mutation,a with high antibacterial activity strain was obtained.Compared with the original strain,its antibacterial effect of Staphylococcus aureus increased by 19.7%,and its antibacterial effect of E.coli increased by 19.5%.The antibacterial activity stays stable after 10 generations of reproduction.
