摘要
采用ISSR标记技术对山西6种天南星科(Araceae)种质资源遗传多样性进行分析。利用NTSYS-pc2.01、POPGEN32软件对扩增结果进行统计和聚类分析,从17个ISSR引物中共检测出146条清晰条带,其中143条为多态性条带,平均多态性位点比率为97.95%,6种种质资源遗传相似系数为0.20~1.00,根据UPGMA法将试验材料聚类为3个类群。结果表明:山西主栽的天南星科资源在分子水平上差异较大;各栽培类型间的差异不仅受遗传因素决定,而且与外界环境因素也有关。遗传多样性的研究可为有效发掘和利用山西天南星科资源提供理论依据。
The genetic diversity of six main Araceae resources in Shanxi was estimated based on inter-simple sequence repeat(ISSR) marker.Genetic similarities were calculated using NTSYS and POPGEN32.At the same time,the dendrogram was constructed using UPGMA method.The 146 highly reproducible ISSR bands were amplified using 17 ISSR primers in which,143 ISSR bands were polymorphic and the percentage of polymorphic loci reached to 97.95%.The genetic similarity coefficient ranged from 0.20 to 1.00.The tested inidivduals were clustered into three groups by using UPGMA cluster analysis.The results showed that Araceae agrads had high genetic diversity,and both genetic and environmental factors contributed to the genetic difference.The research provided theoretical foundation for excavating and utilization of Shanxi Araceae resources.
