摘要
以玉米自交系丹黄25为试材,用农杆菌介导法将玉米淀粉分支酶SBEIIb基因转入玉米中,探讨了农杆菌介导玉米愈伤组织转化的影响因素。结果表明:愈伤组织经过9 d的继代,菌液中乙酰丁香酮(AS)浓度为10 mg/L,侵染时间为20 min,共培养60 h为最佳转化条件。获得的4株再生植株经PCR分析鉴定,其中3株表现阳性,证明外源基因已经整合到玉米基因组中。
The SBEIIb gene was transferred into maize inbred line Danhuang 25 by Agrobacterium tumefaciens mediated transformation. Factors influenced transformation efficiency was studied. The results showed that 9 days with subculture of callus, AS concentration in the bacilli concentration at 10 mg/L, 20 minutes infection time and coculture 60 hours were the optimal conditions. PCR analysis on four resistance plants demonstrated that three of them were positive and the foreign gene was integrated into maize genome.
