摘要
目的检测胃癌组织以及癌旁胃组织中MMP.2、TIMP.2、MMP.9的表达差异,并分析其对胃癌侵袭转移能力的影响及其可能机制。方法将97例胃癌组织以及89例癌旁胃组织制作成组织芯片,用免疫组织化学半定量的方法检测MMP-2、TIMP-2、MMP-9在胃癌组织及癌旁胃组织中的表达程度并分析其与临床病理参数的关系。结果MMP-2、TIMP-2、MMP-9在89例胃癌组织和配对的癌旁胃组织中的表达均有显著差异(P〈O.01),其中MMP-2在胃癌组织的表达大于其在癌旁非肿瘤胃组织,而TIMP.2、MMP.9在胃癌组织的表达小于其在癌旁非肿瘤胃组织。MMP-2的高表达率与胃癌组织的临床分期,浸润深度,淋巴结转移显著正相关(P〈0.01);TIMP.2与胃癌组织的临床分期,浸润深度(P〈0.01),淋巴结转移(P〈0.05)显著负相关。MMP.9在肠型胃癌的表达要显著高于弥漫型胃癌(P〈O.05)。MMP-2的高表达与TIMP-2的高表达显著负相关(P〈O.01)。结论MMP-2可促进胃癌的侵袭转移,在胃癌组织高表达;TIMP-2抑制胃癌的侵袭转移,在胃癌组织低表达;MMP-2的表达与TIMP-2的表达显著负相关。MMP-9在胃癌组织表达低,.在肠型胃癌中的表达显著高于弥漫型胃癌,与TIMP-2的表达无显著相关性。
Objective To detect gastric cancer and adjacent gastric tissue matrix metalloproteinase-2(MMP- 2),tissue inhibitor of metalloproteinase-2(TIMP2),matrix metaUoproteinase-9(MMP-9) expression differences,and analyze the influence for invasion and metastasis of gastric cancer.Methods 97 cases of gastric cancer and 89 cases of adjacent gastric tissue were made into tissue chips,semi-quantitative immunohistochemical method was used to detect the expression of MMP-2,TIMP2,MMP-9 in gastric cancer and adjacent gastric tissues.Results Expressions of MMP-2,TIMP2,MMP-9 in 89 cases of gastric cancer and paired adjacent gastric tissues were significantly different(P 〈 0.0 I)'MMP-2 expression in gastric cancer tissue was greater than that in adjacent non- tumor gastric tissue,and TIMP2,MMP-9 expression in gastric cancer tissue were lower than in adjacent non- tumor gastric tissue.The expression of MMP-2 was significantly positively correlated with the clinical stage of gastric cancer, depth of invasion, lymph node metastasis(P 〈 0.01);and the expression.of TIMP-2 was significantly negatively correlated with the clinical stage of gastric cancer, depth of invasion(P 〈 0.01),lymph node metastasis(P 〈 0.05).The MMP-9 expression in intestinal type gastric cancer was significantly higher than that in the diffuse- type gastric cancer(P 〈 0.05).The expression of MMP-2 and TIMP-2 was significantly negatively correlation(P 〈 0.01).Conclusion Higher expression of MMP-2 in gastric cancer may contribute to invasion and metastasis of gastric cancer, lower expression of TIMP-2 in gastric cancer may inhibit the invasion and metastasis of gastric cancer, MMP-2 expression and TIMP-2 expression was significantly negatively correlated.The expression of MMP-9 in gastric cancer tissues was significantly higher in intestinal type gastric cancer than in diffuse-type Kastric cancer, and had no significant correlation with TIMP2 expression.
