摘要
目的为了辅助筛选高产白藜芦醇的花生毛状根及其愈伤组织株系,选择适合于花生毛状根及其愈伤组织中白藜芦醇的提取方法。方法分别采用有机溶剂提取法、碱提取法和超声波法,提取花生毛状根及其愈伤组织中的白藜芦醇,利用HPLC法测定各提取方法的提取液中白藜芦醇的含量。结果 3种方法中,以超声波法的提取率最高,并且超声30min即获得较好效果。结论超声波法适合于花生毛状根及其愈伤组织中白藜芦醇的提取,有助于辅助筛选高产白藜芦醇的花生毛状根及其愈伤组织株系。
Objective To assist in screening high-yielding resveratrol of peanut hairy roots and calluses lines and to select the extraction method for resveratrol from hairy roots and callus of peanut. Methods To extract resveratrol of peanut hairy root and callus by using organic solvent extraction,alkaline extraction and ultrasonic extraction,and measure the resveratrol contents by HPLC. Results Among the three methods,ultrasonic extraction method has the highest rate,and get better results in 30min ultrasonic extraction. Conclusion The ultrasonic method is suitable for resveratrol extract from the hairy roots and calluses of peanut.It can helps select the high-yielding hairy roots and callus lines of peanut.
