虎杖中白藜芦醇的提取纯化工艺研究 被引量:5

摘要 目的:考察虎杖中白藜芦醇的最佳提取纯化工艺。方法:利用单因素试验对虎杖中白藜芦醇的提取条件,即碱的种类、碱料比、纤维素酶种类、酶料比及酶解时间进行了考察,并选取后4种因素设计了正交试验来优化提取条件;HPLC测定含量;沉淀法纯化白藜芦醇粗品。结果:最佳提取工艺为碱料比(1∶10),用1.5万单位.g-1固体纤维素酶,酶料比(1∶100),酶解3h;沉淀法的较优工艺:80%乙醇为溶剂,乙醇与水的比例为8∶7。结论:此法操作简单易行。 Objective: To study the optimum extraction and purification process of resveratrol from polygonum cuspidatum. Methods: The extracting technology of resveratrol from polygonum cuspidatum was discussed with the mono-factor design and the effects of the kind of alkali, the ratio of the alkali to the solid material, the kind of cellulase, the ratio of the cellulase to the solid material and enzymolysis time were discussed. The later four factors were chosed in the orthogonal design. The determination method of resveratrol content was HPLC. And the proper process of purification of crude product by precipitation was discussed. Results: The optimum extract conditions are as follows: the ratio of the alkali to the solid material is 1:10, using 15 000 unit·g^-1 solid cellulase, the ratio of the cellulase to the solid material is 1:100, enzymolysis time is 3h. The proper conditions of precipitation are 80% ethanol as solvent, the ratio of the ethanol to water is 8:7. Conclusion: The procedure is simple.
