摘要
目的考察祛瘀清热颗粒(大黄、桃仁、桂枝、山楂及甘草)指纹图谱的最适进样量。方法色谱条件Hypersil BDSC18色谱柱(250 mm×4.6 mm,5μm),乙腈-水(0.1%磷酸)梯度洗脱,体积流量1.0 mL/min,检测波长273 nm,柱温30℃。以单峰面积对总峰面积之比设定为信息熵,计算不同进样量的各指纹图谱的信息熵。结果当进样量为15μL时,指纹图谱所得的信息熵最大。即芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚含有量分别为0.032、0.041、0.030、0.084、0.023μg。结论信息熵法可以确定指纹图谱的最佳进样量。
AIM To establish the optimal sample size for the chemical fingerprint of Quyu Qingre Granules(Rhei Radix et Rhizoma,Persicae Semen,Cinnamomi Ramulus,Crataegi Fructus,Glycyrrhizae Radix et Rhizoma).METHODS The HPLC was performed on a Kromasil C18 analytical column(250 mm×4.6 mm,5 μm),and acetonitrile-phosphoric acid water solution(0.1%) was taken as the mobile phase at the flow rate of 1.0 mL/min and the detection wavelength was at 273 nm.The column temperature was maintained at 30 ℃.Ratio of single characteristic peak to total peak in HPLC was selected as the entropy of information and connected with the sample size.RESULTS Fifteen microliter of sample size was calculated as the maximum entropy,which was equivalent to 0.032 μg aloe-emodin,0.041 μg rhein,0.030 μg emodin,0.084 μg chrysophanol and 0.023 μg physcion.CONCLUSION Entropy of information of fingerprint can determine the optimal sample size of Quyu Qingre Granules.
