正交设计法优化大鼠肝细胞色素P450的孵育体系 被引量:2

摘要 目的:考察大鼠肝细胞色素P450 1A2,2A6,2C9,2E1,3A2 5种亚型的最佳体外孵育条件。方法:通过正交试验,以对乙酰氨基酚、7-羟基香豆素、4-羟基甲苯磺丁脲、6-羟基氯唑沙宗、6-β羟基睾酮总含量为考察指标,优化大鼠肝细胞色素P450的体外孵育条件。结果:大鼠肝细胞色素P450最佳体外孵育条件为温度37℃,孵育时间90 min,酶含量0.6 mg.mL-1。结论:优选的体外孵育条件为研究经P450 1A2,2A6,2C9,2E1,3A2代谢的药物相互作用及其他物质对该酶的影响提供依据。 OBJECTIVE To optimize the incubated conditions of eytochrome P450 1A2,2A6,2C9,2E1 and 3A2 of rats in vitro. 1VIETttODS The incubated conditions were optimized by orthogonal design with the content of acetaminophen, 7- hydroxycoumarin,4-hydroxytolbutamide, 6-hydroxychlorzoxazone and 6β-hydroxytestosterone as index, RESULTS The opti- mum incubated conditions were as follows: temperature 37 ℃; incubation time 90 min; liver microsomes 0. 6 mg·mL^-1. CON- CLUSION The incubated conditions of cytochrome P450 in vitro can be applied for studying on the interaction between drugs metabolized by cytochrome P450 1A2,2A6,2C9,2E1 and 3A2.
