摘要
利用LiCl和紫外线诱变圆红冬孢酵母AS 2.1389,以红四氮唑(TTC)为显色剂通过96孔板进行初筛;通过气相色谱(GC)进行复筛,得到突变株D2。与原始菌株相比,D2的脂肪酸含量提高了72%,α-亚麻酸(GLA)含量提高了259%,达到0.508g/L。经传代培养,其GLA产量稳定。
The original strain Rhodospridium toruloides AS 2.1389 was treated by LiC1 and UV. The primary screening was carried out by using TTC as a color reagent with 96 orifice plate, and the secondary screening was detected by GC method, eventually a high-yielding alpha-linolenic acid mutant strain D2 was obtained. The results showed that the lipid content of mutant sWain was increased by 72% and the alpha-linolenic acid content was 2.59 times higher than that of the original strain, which was up to 0.508g/L. After subculturing, the 8^th generation was stable heredity.
