摘要
为提高林荫银莲花(Anemone flaccida Fr.Schmidt)不定芽繁殖系数,对其组培快繁体系进行优化。以林荫银莲花无菌苗为外植体,研究了培养基中激素、碳源、添加物和矿质元素钙等组分对其不定芽增殖和生长状况的影响,得出最佳的不定芽增殖培养基配方。以1/2 Ca浓度MS为固体培养基,激素为1.0 mg/L 6-BA和1.0 mg/L NAA,蔗糖或食用白糖30 g/L,水解酪蛋白为添加物,浓度为750 mg/L,培养30 d后繁殖系数可达4.01,大幅提高了不定芽的增殖速度和质量,为工厂化生产林荫银莲花种苗奠定了基础。
To improve the adventitious bud multiplication coefficient of Anemone flaccida Fr.Schmidt,the kinds and doses of some components in the multiplication medium were adjusted.The aseptic seedlings were used as explants and cultivated in MS solid medium,and the influences of some medium components,such as hormone composition,carbon source,organic additive content and calcium content,on adventitious bud multiplication and growth were investigated.The results showed that MS(1/2 Ca) supplemented with 1.0 mg/L 6-BA and 1.0 mg/L NAA,30 g/L sucrose or edible sugar as carbon source and added with casein hydrolysate 750 mg/L,was suitable for proliferation of A.flaccida,in which multiplication coefficient can be achieved 4.01 times after culturing for 30 d.This study optimizes tissue culture and regeneration system of A.flaccida by increasing the proliferation of adventitious bud speed and quality,thus laying a basic foundation for commercial producing large-scale seedlings.
