摘要
注射hCG18~19 h 后收集的小鼠卵母细胞, 85 % 以上可为乙醇所激活, 产生均质单倍体、嵌合单倍体、1 个原核的杂合二倍体和2 个原核的杂合二倍体4 种激活类型, 且它们的比率分别为81-4 % 、10-5 % 、4-6 % 和3-5 % 。单倍体胚胎体外培养后有少数可发育至囊胚期。单倍体孤雌胚在8细胞期开始出现二倍体核型, 从而形成单倍体胚胎、单倍体二倍体嵌合胚和二倍体胚胎。孤雌胚的核型中还出现较高比率的非整倍体。
The mouse eggs were activated by ethanol,and the development and karyotypes of the resulting parthenogenetic embryos were investigated.More than 85% of oocytes could be activated by ethanol treatment and four types of parthenogenetic activation(uniform haploid,mosaic haploid,heterozygous diploid with one pronucleus and heterozygous diploid with two pronuclei)were produced,and the rate of them were 81 4%、10 5%、4 6% and 3 5% respectively.Haploid embryos could develop to blastocyst stage in vitro,but the rate was low.Diploid karyotypes appear at 8 cell stage in parthenogenetic morula derived from haploid activated oocytes.Out of twelve 8 to 16 cell parthenogenones,8 specimens displayed haploidy,3 haploidy and diploidy,and 1 possessed only diploid metaphase.The results of karyotype analysis also showed that 6 8% was aneuploid out of parthenogenetic morulae.
