摘要
目的 探讨铅对人神经胶质瘤细胞(U251)中蛋白激酶C(PKC)的mRNA和蛋白表达的影响.方法 以0.05、0.50、5.00、50.00、500.00、900.00和1000.00 μmol/L酪酸铅[Pb(Ac)2]分别处理U251细胞24 h,噻唑蓝(MTT)法检测铅对U251细胞的毒性;以0.05、5.00、500.00 μmol/L Pb(Ac)2分别处理U251细胞24 h,用反转录-聚合酶链反应(RT-PCR)法和蛋白免疫印迹法(Western blot)检测PKC的mRNA和蛋白相对表达水平.结果 与对照组(100%)比较,5.00、50.00、500.00、900.00和1000.00 μmol/LPb( Ac)2处理U251细胞24 h后细胞存活率明显下降,分别为84.5%、78.2%、76.5%、50.3%和43.2%,差异有统计学意义(P<0.01),且呈剂量-反应关系(rs=-0.947,P<0.01),随着染毒剂量的增加,细胞存活率下降.500.00 μmol/L Pb(Ac)2处理细胞24 h,PKC mRNA相对表达量降低为0.40±0.01,与对照组(0.51±0.02)比较,差异有统计学意义(P<0.01).0.05、5.00、500.00 μmol/L Pb (Ac)2处理U251细胞24 h可使PKC蛋白相对表达量降低,分别为0.68±0.02、0.62±0.01和0.33±0.02,与对照组(0.98±0.01)比较,差异有统计学意义(P<0.01).结论 铅使U251细胞存活率下降,细胞中PKC的mRNA和蛋白表达下调.
Objective To observe the effects of lead on mRNA and protein expression of PKC in U251 cell line.Methods After U251 cells were exposed to 0.05,0.50,5.00,50.00,500.00,900.00 and 1000.00 μmol/L Pb(Ac)2 for 24 hours,the cytotoxicity of Pb on U251 cells was measured by MTT assay.RT-PCR and Western blot assay were used to detect the mRNA and protein expression levels of PKC in U251 cells exposed to 0.05,5.00 and 500.00 μmol/L Pb (Ac)2 for 24 hours.Results The survival rates of U251 cells treated with 5.00,50.00,500.00,900.00 and 1000.00 pmol/L Pb (Ac)2 were 84.5%,78.2%,76.5%,50.3% and 43.2%,respectively,which were significantly lower than those of control group (P〈0.01).The PKC mRNA expression level (0.40±0.01) of U251 cells treated with 500.00 μmol/L Pb (Ac)2 was significantly lower than that (0.51±0.02) of control group (P〈0.01).The PKC protein expression levels of U251 cells treated with 0.05,5.00 or 500.00 μmol/L Pb(Ac)2 were 0.68±0.02,0.62±0.01 and 0.33±0.02,respectively,which were significantly lower (0.98±0.01) than those of control group (P〈0.01).Conclusion Lead can decline the cell viability,PKC mRNA and protein expression levels of U251 cells.
