摘要
目的:建立异丙肾上腺素(Iso)诱导的大鼠心肌肥厚模型,观察大豆苷元(DD)对大鼠心肌肥厚时炎性介质的影响。方法:健康雄性SD大鼠随机分为5组:对照组、心肌肥厚模型组、溶剂对照组、低剂量(0.1μmol/kg)及高剂量(0.3μmol/kg)DD组。各组分别背部皮下注射同体积NS或Iso 1 mg/kg,每日一次,连续10 d,建立心肌肥厚模型。连续皮下给药14 d。称体重及左心室重量并计算左心室重量指数,化学比色法检测血清MPO活性,ELISA方法检测心肌组织IL-2及TNFα含量。结果:与对照组相比,模型组左心室重量指数、血清MPO活性、心肌组织IL-2及TNFα水平升高;与模型组相比,DD组左心室重量指数、血清MPO活性、心肌组织IL-2及TNFα水平降低。结论:DD对异丙肾上腺素诱导的大鼠心肌肥厚具有保护作用,干预炎性介质的生成是DD抗心肌肥厚的机制之一。
Objective:To explore the anti-inflammation effects of daidzein on rat's myocardial hypertrophy induced by isopropylarterenol(Iso).Method: The rat's myocardial hypertrophy model is formed through hypodermic injection which extended 10 days at back skin with Iso(1 mg/kg·d),it will be administrated with DD(0.1or 0.3 μmol/kg·d),DMSO or NS in subcutaneous injection way from the second day,and the process will extend 14 days.After the last administration,the rat will be in absolute diet status for 12 hours and its blood and heart are taken under anesthesia.The indexes of left ventricle will be calculated.The level of IL-2 and TNFα in myocardial tissue and the activity of MPO in serum will be investigated.Result:It show that the index of left ventricle of Iso group are much bigger than control one,the activity of MPO are increased in serum,and the level of IL-2 and TNFα in myocardial tissue is enhanced.However,these are inverted after treat with DD.Conclusion:DD has a protection effect on rat's myocardial hypertrophy induced by Iso because it decreases the inflammation mediators in rats.
