摘要
目的:制备别嘌醇缓释微丸,考察其释药机制。方法:采用正交试验法进行处方筛选优化,用数学模型拟合释放曲线。结果:微晶纤维素(MCC)的用量对微丸的成球性、释药速率有显著的影响,别嘌醇缓释微丸的体外释药行为符合Higuchi方程释放模型。结论:制备别嘌醇缓释微丸具有较好的缓释效果。
Objective: To prepare allopurinol sustained-release pellets and investigate the drug release mechanism. Method: An orthogonal experiment was designed to Optimize the formulation of the allopurinol sustained-release pellets and a mathematical model was fitted to the release profiles. Result: The effect of microcrystalline cellulose (MCC) amount on the pellet formation and release was significant. The in vitro release behavior of the prepared allopurinol sustained-release pellets was fitted to the Higuchi release model. Conclusion: The allopurinol sustained-release pellets show promising sustained-release property in vitro.
