迄今研究高等植物的β-甘露聚糖酶主要是用贮藏甘露聚糖的种子为材料(Halmer等1975,Halmer和Bewley 1979,Reid等1977,McCleary 1983)。营养器官贮藏甘露聚糖的植物,仅存在于单子叶植物的少数科(Meier和Reid 1982)。魔芋球茎贮藏大量甘露聚糖,Sugiyama等(1973)和Shimahara等(1975)
The changes of β-mannanase activities in the corms of Amorphophallus konjac during dormancy, growth and development have been investigated, and a β-mannanase was partial purified from the dormant corms by chromatography with DEAE-Cellulose (DE-52) and gel-filtration on Sephadex G-100. The enzyme had an optimal pH of 5.3 and optimum temperature of 40℃, and was quite stable at pH 4.0~8.6. The values of K_m and V_(max) for A. konjac β-mannanase was 4.2×10^(-2) (%) and 9.1×10^(-3) (△OD_(560nm) min^(-1) ml^(-1)), respctively, Mannan hydrolysis by this enzyme was by typical random mechanism.
