离子交换树脂固定化脂肪酶催化合成蔗糖乙酯 被引量:4

Enzymatic synthesis of sucrose acetate by immobilized Candida rugosa lipase on ion exchange resin

摘要 南极假丝酵母(Candida rugosa)脂肪酶具有优良的催化性能,对其进行固定化可以很方便地实现酶的回收和再利用.采用4种离子交换树脂为载体,对来源于南极假丝酵母的脂肪酶进行了吸附固定化,并对固定化酶催化合成蔗糖乙酯的活性进行考察.结果表明,以弱碱性丙烯酸系阴离子交换树脂214为载体的固定化酶活性最高.固定化酶制备过程中缓冲液的最适宜pH值为8.4,最佳固定化时间为3h,最佳酶液浓度为8mg/mL,最适固定化温度为35℃.所得固定化酶催化合成蔗糖乙酯,酯化率达57.12%,蔗糖-6-乙酯的选择性达到51.21%.10批反应后,该固定化酶仍有较高的酶活力. To easily recycle and reuse Candida rugosa lipase with excellent catalytic properties,4 types of ion exchange resins were screened as the carriers to immobilize the Candida rugosa lipase by adsorption.And immobilized enzyme catalyzed transesterification of sucrose with vinyl acetate was investigated.The results showed that the lipase immobilized on anionic resin 214 had the highest specific activity.The optimal conditions of immobilization were as follows: buffer pH=8.4,adsorption time 3 h,concentration of Candida rugosa lipase 8 mg/mL and temperature 35 ℃.The esterification rate of sucrose acetate reached 57.12%,the selectivity of 6-sucrose acetate reached 51.21%.The activity of immobilized enzyme kept high after 10 cycles of reactions.
