摘要
目的观察吉米沙星对肺炎支原体(Mp)诱导A594细胞产生IL-8和TNF-α的影响,为临床治疗肺炎支原体肺炎提供理论依据。方法培养A549细胞和Mp,用不同感染复数的Mp刺激A549细胞,同时加入不同浓度的吉米沙星与之共同孵育,ELISA检测IL-8和TNF-α的分泌情况。结果 Mp能诱导A549细胞产生IL-8和TNF-α,1～20mg/L吉米沙星对其具有抑制作用,但随着Mp感染复数的增加,该抑制效应降低。结论吉米沙星可抑制Mp诱导的A549细胞分泌IL-8和TNF-α,提示吉米沙星在治疗Mp感染当中可能具有抗菌、抗炎的双重作用。
Objective This study was designated to investigate the effect of Gemifloxacin on IL-8 and TNF-ct production in A549 cells induced by Mycoplasma pneumoniae. Methods A594 cells and M. pneumoniae were co-cultured in vitro, different concentration of Gemifloxacin were added into A594 cells which were infected by M. pneumoniae at different multiplicity of infection （MOI）. The level of IL-8 and TNF-α were detected by ELISA. Results M. pneumonie could induce IL-8 and TNF-α production in A549 cells. The contents of IL-8 and TNF-α were decreased after co- cultured with 1-20mg/L of Gemifloxacin. The inhibitory effect was attenuated with the increase of MOI of M. pneumoniae. Conclusion Genifloxacin can inhibit the production of IL-8 and TNF-α, which indicated that Gemifloxacin may have dual effects of antibiosis and anti-inflammatory.
