新疆地区维吾尔族寻常型银屑病与HLA-DRB1~* 07等位基因的关联研究 被引量:6

摘要 目的银屑病是慢性炎症性、增生性皮肤病,可分为Ⅰ型和Ⅱ型2种亚型,发病机制尚不清楚,与遗传、免疫、感染等因素有关。近年来,国内外对银屑病发病与人类白细胞抗原(human leucocyte antigen,HLA)相关性的研究积累了一些资料。目前认为HLA-DRB1*07是与寻常型银屑病最为密切相关的基因位点。尚未见有维吾尔族银屑病患者HLA-DRB1*07频率的研究报道。文中探讨新疆地区维吾尔族寻常型银屑病与HLA-DRB1*07等位基因的相关性。方法用聚合酶链反应-序列特异性引物(polymerase chain reaction-sequence specific primer,PCR-SSP)对200例维吾尔族寻常型银屑病患者HLA-DRB1*07等位基因进行检测,并与200例健康人群进行对照,分析了携带该基因的银屑病患者与其家族史的相关性。结果寻常型银屑病患者组HLA-DRB1*07等位基因频率显著高于对照组。Ⅰ型寻常型银屑病HLA-DRB1*07等位基因频率显著高于对照组。有银屑病家族史患者与无银屑病家族史的患者HLA-DRB1*07等位基因频率无显著性差异。结论 HLA-DRB1*07等位基因可能是新疆维吾尔族寻常型银屑病患者的易感基因。 Objective Psoriasis is a chronic inflammatory hyperplastic skin disease,whose pathogenesis is not yet clear though known to be associated with genetic,immune and infectious factors.In the recent years,progress has been made in the studies of the relationship between psoriasis and human leucocyte antigens(HLA).HLA-DRB1*07 is one of gene sites most closely related with psoriasis.However,no studies have been reported on the frequency of HLA-DRB1*07 in psoriasis patients in the Uygurs of Xinjiang area.Our aim is to investigate the correlation of psoriasis vulgaris(PV) with the HLA-DRB1*07 allele in Xinjiang Uygurs.Methods We detected the distribution frequencies of the HLA-DRB1*07 allele in 200 PV patients in Xinjiang Uygurs by polymerase chain reaction-specific sequence primers(PCR-SSP),compared them with 200 healthy controls,and analyzed the correlation of the gene carriers with their family history.Results The frequency of the HLA-DRB1*07 allele was obviously higher in the PV patients than in the controls,and so was that of the type-I PV patients.No significant differences were found in the frequency of the HLA-DRB1*07 allele between the PV patients with positive history and those without.Conclusion The HLA-DRB1*07 allele may be a predisposing gene of psoriasis vulgaris in Xinjiang Uygurs.
