摘要
目的:探讨低剂量辐射(low dose radiation,LDR)对瘤细胞蛋白激酶α(PKCα)的影响。方法:昆明种雄性小鼠100只随机分为对照组、LDR1-4组。左后肢腹股沟皮下接种S180肉瘤细胞。LDR1-4组接种后11 d予γ射线全身照射75 mGy,照射后8、12、24、48 h处死。对照组在24 h被处死。免疫组化法检测PKCα和TERT的浓度。结果:与对照组比较,LDR1组差异无统计学意义,P>0.05。LDR2-4组PKCα的浓度降低,且LDR3组降到最低。结论:低剂量辐射能减少瘤细胞PKCα的分泌,从而抑制瘤细胞的生长,且照射后24 h降到最低。
Objective:To study the effect of low dose radiation(LDR) on protein kinase PKCα in tumor cells.Methods:Male Kunming mice were randomly divided into five groups: direct tumor-bearing group(DT group) and low dose radiation 1-4 groups.The mice were subcutaneously implanted with S180 sarcoma cells in the left hind legs.On 11 d after the implantation,the mice of LDR 1-4 groups were given 75 mGy whole body γ-ray radiation and sacrificed after 8,12,24,48 h.DT group was sacrificed at 24 h after γ-ray radiation.The PKC α levels of tumor cells were analyzed by immunohistochemistry.Results:Compared with DT group,the expression of PKCα was significantly reduced in the LDR2-4 groups,the lowest in the LDR3 group and that in LDR1 showed no statistical difference.Conclusion:Low dose radiation of 75 mGy can decrease the production of PKCα.
