癌胚抗原阴性的人大肠癌组织蛋白质组的研究 被引量:1

摘要 目的:比较癌胚抗原(CEA)阴性的人大肠癌组织与CEA阳性的人大肠癌组织之间的蛋白质表达差异。方法:收集大肠癌患者切除癌组织的新鲜标本,根据血清CEA分为阴性组、阳性组,同时取正常黏膜作对照组,提取各组组织的总蛋白,进行双向凝胶电泳和凝胶图谱差异分析,比较各组组织之间表达差异点,将这些差异蛋白点进行质谱分析数据库鉴定。结果:各组双向电泳胶上蛋白斑点图象清晰,阴性组和阳性组中差异表达的蛋白点18个,成功鉴定其中10个为9种蛋白质。其中,在阴性组中特异表达的蛋白质点为含波形蛋白的混合物1、肝脂肪酸结合蛋白,表达上调的蛋白质点为肌动蛋白结合蛋白22-α、热休克蛋白27序列、血红蛋白β链突变体;在阳性组中特异表达的蛋白质点磷酸甘油醛异构酶、角蛋白10、角蛋白1,表达上调的蛋白质点为β微管蛋白链。结论:CEA阴性的人大肠癌组织与CEA阳性的人大肠癌组织存在蛋白质组差异表达,这些蛋白可能与大肠癌表达CEA有关。 Objective:To evaluate the proteomic differences among the CEA negative and positive human colorectal carcinoma tissues. Methods:Fresh human colorecta] carcinoma tissues were collected and classified into CEA-negative group and CEA-positive group according to the serum CEA level. Tissues of normal mueosa were enrolled as control group. Proteins in the three groups was separated, and then processed by two- dimensional gel electrophoresis (2-DE). Spots with differential expression were identified by mass spectrometry. Results: Distinct images of protein spots on 2-DE gel in each group were obtained. There were 18 differentially expressed protein spots in CEA negative and positive groups, 10 of which were successfully identified to be of 9 types of protein. In CEA-negative group, spots of vimentin-containing mixture I and liver fatty acid-binding protein were differentially expressed, and those of actin binding protein 22-a, HSP27 sequences and mutant hemoglobin beta chain were upregulated. In CEA positive group, spots of triosephosphate isomerase, keratin 10 and keratin 1 were differentially expressed, and those of tubulin beta chain were upregulated. Conclusion: Differentially expressed proteins probably correlated with CEA expression were identified between CEA negative and positive human colorectal carcinoma tissues.
