小檗碱诱导体外培养PC3细胞凋亡的作用 被引量：7

摘要 目的观察小檗碱对体外培养人前列腺癌PC3细胞凋亡的作用。方法体外培养前列腺癌PC3细胞,分别用10、20和40μmol/L小檗碱处理细胞后,采用MTT法检测其对细胞增殖的影响,20μmol/L小檗碱处理细胞后流式细胞术检测细胞周期,吖啶橙染色法检测细胞凋亡情况,并通过Western印迹法检测Bcl-xL及前列腺特异性抗原(PSA)蛋白表达。结果小檗碱处理后,PC3细胞出现增殖抑制,细胞周期停滞于G2/M期,吖啶橙染色呈黄染,PSA及Bcl-xL蛋白表达明显下调。结论小檗碱可诱导人前列腺癌PC3细胞凋亡,其作用机制可能与Bcl家族信号通路有关。
