摘要
目的:研究四逆散及其不同配伍对实验性UC大鼠NF-κB和IL-1βmRNA表达的影响。方法:将实验动物分为14组,免疫法造模,采用Q-PCR技术检测实验大鼠结肠组织NF-κB和IL-1βmRNA的表达。结果:模型组NF-κB mRNA表达(2.22±0.44)明显高于正常组(1.00±0.26),P<0.01;枳甘组(4.66±0.81)显著高于模型组(P<0.01),其余各组均显著低于模型组(P<0.01或P<0.05);与正常组(1.00±0.27)比较,模型组、柴枳甘组、芍甘组和枳甘组实验大鼠结肠黏膜IL-1βmRNA表达水平明显升高,P<0.01;与模型组(7.84±1.18)比较,各药物组实验大鼠结肠黏膜IL-1βmRNA表达水平均显著降低,P<0.01。结论:四逆散能够通过下调结肠黏膜NF-κBI、L-1βmRNA的基因表达干预UC,方中柴胡、芍药发挥了君药、臣药的重要作用,并体现为相须配伍;就四逆散下调UC大鼠结肠黏膜IL-1βmRNA的表达的作用而言,方中起主要作用的是芍药。
Abstract:Objective:To study the Sini powder and the different compatibilities in adjusting NF - κB and IL - 1β mRNA expressions of experimental UC rats. Methods:The experimental animals were divided into 14 groups, immunological modeling, using Q -PCR detection to determine experimental rat colon the expressions of NF- κB and IL- 1βmRNA. Results :The NF- κB mRNA expression(2.22 ± 0.44) was significantly higher than that of the normal group ( 1.00 ± 0.26), P 〈 0. 01 ;Zhi gan group (4. 66 ± 0. 81 ) was significantly higher than the model group( P 〈 0. 01 ), the remaining group was significantly lower than model group(P 〈0.01 or P 〈0.05); with the normal group( 1.00 ±0. 27) compared,the IL- 1βmRNA expression of model group, Chaizhigan group, Shaogan group and Zhican group were significantly higher, P 〈 0.01 ; with model group (7.84 ±1.18 ) compared, each drug group was all significantly lower, P 〈 0.01. Conclusion:Sini Powder can lower NF- κB and IL- 1β mRNA gene expressions to interfere UC. Bupleurum and Peony played the important roles.
