30多年前在果蝇体内鉴定到了PcG(Polycomb group)蛋白复合体,它是通过染色质修饰调控靶基因转录的表达抑制因子,通过抑制同源异形基因(homeotic gene,Hox)的表达调控机体发育。其相关蛋白质RYBP(Ring1 and YY1 binding protein)分布于细胞质和细胞核,参与调节基因转录、个体发育、细胞凋亡以及人类恶性肿瘤的发生等生物学过程。本文就RYBP的特性、作用机制及生物学功能进行综述。
Polycomb group (PcG) proteins are transcriptional repressors that epigenetically modify chromatin, and they were identified in Drosophila more than 30 years ago. PcG can regulate anterior-posterior body patterning through the repression of Hox genes. PcG- associated protein RYBP (Ringl and YYl binding protein) is expressed both in cytoplasm and in nuclear. Studies revealed that RYBP protein was involved in repressing transcription, inducing cell apoptosis, correcting execution of developmental programs and human tumorigenesis. This paper summarizes the recent studies on Polycomb group, the character of RYBP, its molecular function mechanisms and its biological functions.
