冬虫夏草含药血清诱导神经干细胞分化的研究

摘要 目的:研究神经干细胞具有自我更新、未分化和多向分化潜能,冬虫夏草中的氨基酸,核苷和糖蛋白等能诱导神经干细胞的定向分化。方法:取新生24h内的W istar仔鼠海马区分离培养神经干细胞三代后,加入低,中,高浓度冬虫夏草含药血清与对照血清进行培养,观察其对神经干细胞影响,免疫组化法检测神经干细胞向神经元分化的情况。结果:新生大鼠海马分离培养的神经克隆球,经免疫细胞化学染色检测为N estin阳性细胞。N SC贴壁分化的细胞呈N SE、GFAP阳性。冬虫夏草低,中药物实验组诱导神经干细胞分化为神经元的阳性细胞明显高于对照组,且呈量效依赖关系,高浓度药物实验组与对照组没有明显差别。结论:冬虫夏草药物血清可体外诱导神经干细胞向神经元方向分化,在一定范围内存在量效依赖关系。 Objective.. To study the potential of the self--renewing,undifferenfiation and multi--differenti- ation of neural stem cells from Cordyceps sinensisin and the direct differentiation induced by its amino acids, nu- cleotides and sugar protein. Methods:The newborn of the Wistar rat hippocampus within 24 hours was isolated and neural stem cells of three generations were cultured, and after that,low, medium and high doses of Cordyceps sinensis containing serum and control serum culture medium were added for the observation the effects on its neural stem cells and the differentiation of the neural stem cells into neurons was detected by immunohistochemistry and semi-quantitative RT--PCR detection. Results: The isolated hippocampal nerve clone ball, showed Nestin -- positive cells by immunocytochemistry. After the Brdu incubation, the newly formed neuro- spheres were marked as BrdU -- positive immune. The differentiated cells adhering to NSC were NSE, GFAP positive. Cordyceps sinensis was low, the positive cells differentiated by the experimental group of drug induced neural stem cells were significantly higher,and its effect depended on the dose, with no significant difference be- tween experimental group and control group at the high concentrations of drugs. Conclusion.The serum Cordy- ceps can induce neural stem cells to differentiate into neurons, and to some extent, the effects depend on the dose.
