内源性硫化氢与皮质发育障碍模型大鼠认知功能损害的相关性研究

Relationship between Cognitive Impairment and Endogenesis Hydrogen Sulfide in Rats with Disorders of Cortical Development

摘要 目的探讨皮质发育障碍DCDs(disorders of cortical development)模型鼠的认知功能与H_2S的变化关系。方法用γ射线照射孕15d Wistar大鼠,制作DCDs模型。采用Morris水迷宫法测试P30(出生后30天)、P60、P90 F1代DCDs模型鼠和对照组大鼠的学习能力和空间记忆能力。用敏感硫电极法检测大鼠血液及海马组织中H_2S浓度。用亚甲基蓝法测定大鼠海马神经元细胞胱硫醚-β-合酶CBS(cystathionine-beta-synthase)酶活性。结果水迷宫实验:P60、P90模型组大鼠较正常对照组潜伏期明显延长(P<0.05),且随月龄增加潜伏期越长。P60、P90模型组大鼠较对照组血液H_2S浓度分别降低6.5、11.5μmol/L、海马组织H_2S含量分别减少3.2、5.6μmol/L.mg,海马组织神经元细胞CBS酶活性降低24.94、34.46μmol/L.g.min。P30模型组与对照组的各项变化不明显。结论 DCDs模型鼠脑内H_2S含量及海马组织神经元细胞CBS酶活性降低,内源性H_2S降低与DCDs认知功能损害可能具有相关性,并为治疗皮质发育障碍的认知功能损害可能提供治疗途径。 Objective To explore the changing relations between the cognitive function of cortical developmental disorders (DCDs) model rat and H2S. Methods Animal models in DCDs are "y-radiated pregnant rats. The rats were divided into model groups and normol control groups. (1) The memory capacity was tested by using a Mort'is-water-maze method. (2) The H2S concentration was deteeted in rats blood and the hippoeampus by sensitive sulfur electrode. (3)The CBS (eystathionine-beta-synthase) enzyme activity in the rat hippocampal neuronal cell was detected by methylene blue method to determine. Results ( 1 ) The searching platform latency of model rats was significantly longer than that of the normal controls. (2) The blood H2 S concentration in the P60,P90 months model group rats than in the normal eontrol group lower respeetively 6.5,11.5 μmol/L, the H2 S eoneentration of hippoeampus organization decreased by 3.2 and 5.6 respectively μmol/L, m g, the CBS enzyme aetivity of organization neurons redueed by 24.94,34.46 μmol/L, g. rain. (3)The change is not hange signifieantly in the P30 model group and the normal control group. Conclusion The blood H2S concentration,the H2S concentration of hippocampus organization and the CBS enzyme activity of organization neurons in DCDs model is reduced ,endogenous H2S reduction and DCDs cognitive impairment may have a correlation, and for the treatment of cognitive impairment in the cortex of developmental disorders may provide therapy method.
