摘要
目的探讨出生前暴露于1 800 MHz电磁波对大鼠海马胶原纤维酸性蛋白(GFAP)表达的影响。方法 7周龄SD雌性大鼠32只,雄性16只,适应环境一周后交配(雌∶雄=2∶1),受孕后雌鼠随机分为2个暴露组和2个对照组。暴露组均采用1 800 MHz连续射频电磁波于孕0~20 d进行21 d全身暴露,每天12 h,功率密度分别为0.5和1.0mW/cm2,各暴露组分别设置虚拟暴露作为对照组。暴露完成后在仔鼠长至3周龄和7周龄时每组随机抽取8只,采用免疫组化法对海马GFAP进行染色,采用图像分析系统分析海马CA1、CA3和DG区(齿状回)GFAP的表达,用平均光密度(MOD)值表示。结果未发现1.0 mW/cm2电磁波暴露对3周龄、7周龄子代大鼠海马CA1、CA3和DG区GFAP的表达有影响,其表达与对照组差别无统计学意义。0.5 mW/cm2暴露组3周龄子代大鼠海马各区GFAP表达和对照组差别无统计学意义;0.5 mW/cm2暴露组7周龄子代大鼠海马CA1、CA3和DG区GFAP表达MOD值分别为0.052,0.049,0.053,与对照组7周龄子代大鼠海马各区GFAP表达MOD值(0.106,0.097,0.106)相比表达下调,经两两比较,差异有统计学意义(P<0.05)。结论本实验条件下,出生前暴露于频率1 800 MHz、功率密度0.5 mW/cm2的电磁波可使7周龄大鼠海马CA1、CA3和DG区GFAP表达下调。
Objective To identify the effects on expression of GFAP in rat hippocampus after prenatal exposure to 1 800 MHz radiofrequency field. Methods A total of 32 SD female rats aged 7 weeks were randomly divided into four groups(eight rats in each) after one week of environmental adaptation and successful mating. The rats in the exposure groups were respectively and continuously radiated between 0 days of pregnancy and 20 days of pregnancy (12 hours/day) by 1 800 MHz microwave fields with power density of 0.5 mW/cm^2 or 1.0 mW/cm^2. Simultaneously, two sham-controls were carried out respectively. When the offspring were three weeks and seven weeks old, eight pups were randomly selected from each group and the brains were collected for immunohistochemistry, computer assisted image analysis system was used for the analysis of the expression of GFAP receptor. Results In the 0.5 mW/cm^2 exposure group GFAP expression decreased significantly in the area of CA1, CA3 and DG of hippocampus in seven weeks compared with the control groups. Conclusion Prenatal exposure to 1 800 MHz, power density of 0.5 mW/cm^2 radiofrequency field may down-regulate the expression of GFAP in the area of CA1, CA3 and DG of hippocampus of seven weeks old rats.
