摘要
目的探讨雷公藤甲素对新生大鼠心肌细胞搏动的影响。方法新生大鼠心肌细胞予0、2.5、5、10、20、40、80、160和200μmol/L雷公藤甲素处理,采用实时细胞分析系统(RTCA)于给药前至给药后20 h连续记录心肌细胞搏动频率的变化。结果浓度大于等于2.5μmol/L的雷公藤甲素于给药后10 min可明显导致心肌细胞搏动频率明显下降,但1h后可基本恢复至给药前水平,给药后1h至给药后10 h,细胞搏动频率未见明显异常变化,心肌细胞搏动节律正常。160和200μmol/L组给药后20 h,细胞搏动频率明显下降,正常节律性搏动消失。结论较高浓度的雷公藤甲素给药后期对大鼠心肌细胞毒性作用显著。
Objective To investigate the effect of triptolide on beating rate of neonatal rat cardiac myocytes. Methods Beating rates of primary cultured neonatal rat cardiac myocytes were monitored by real time cell analyzer (RTCA)for continual 20 hours after giving triptolide at vavious doses (0, 2.5, 5, 10, 20, 40, 80, 160 and 200μmol/L). Results Decreased beating rates were observed at 10 min after administration of triptolide (≥2.5 μmol/L) and these decreased rates were recovered after 1 h. Beating rates and rhythm of cardiac myocytes did not showed any abnormal from 1 h to 10 h after dosing. Significant decreased beating rates of cardiac myocytes were observed at 20 h after dosing of 160 and 200 μmol/L triptolide while normal rhythmic beating was no longer observed. Conclusion Triptolide at higher concentration has the toxic effect on primary cultured neonatal rat cardiac myocytes during the later phase of treatment.
