摘要
目的:观察大黄及其有效成分大黄素对人肺腺癌A549细胞的影响,为临床抗肿瘤提供实验证据。方法:采用实时细胞分析系统和改良MTT方法,观察单味中药大黄(0.05μg/L、0.5μg/L、5μg/L)及其有效成分大黄素(16 nmol/L、80 nmol/L、400 nmol/L、2μmol/L、10μmol/L)对A549细胞的作用,同时观察FRAP/mTOR抑制剂、PI3K抑制剂、MAPKK抑制剂和Caspase-3抑制剂对上述作用的影响。结果:大黄及其有效成分大黄素能显著抑制A549细胞增殖,并呈剂量依赖性,动态IC50研究发现大黄素在55 h时IC 50值最低,反应此时刻抑制A549细胞效应最大,其中Caspase-3抑制剂可以明显阻断大黄素对A549细胞的抑制作用。结论:大黄及其有效成分大黄素能有效抑制人肺腺癌A549细胞的增殖,呈剂量依赖性,证实大黄素是通过激活Caspase-3来抑制肺腺癌细胞的增殖。
Objective:To observe the effect of rhubarb and its active component Rheum Emodin on A549 cells in adenocarcinoma of lung,and provide the experimental evidence for clinical practice.Method: With real-time cell analytical system and upgrade MTT,the effects of rhubarb(0.05 μg/L,0.5 μg/L,5 μg/L) and its active component Rheum Emodin(16 nmol/L,80 nmol/L,400 nmol/L,2 μmol/L,10 μmol/L) were observed on A549 cells in adenocarcinoma of lung.At the same time the effects of FRAP/mTOR inhibiter,PI3K inhibiter,MAPKK inhibiter and Caspase-3 inhibiter were also observed on them.Result: Rhubarb and its active component Rheum Emodin can inhibit the A549 cell multiplication,and showed dose dependence.And the dynamic IC50 study demonstrated that the most effective time of Rheum Emodin was at 55th hour.It indicated that at that time A549 cells were restrained the most greatly,and Caspase-3 inhibitor could significantly block the effects of Rheum Emodin on A549 cells.Conclusion: Rhubarb and its active component Rheum Emodin can significantly inhibit A549 cell multiplication in an dose-dependent manner.It is evident that Rheum Emodin inhibits A549 cell multiplication through activating caspase-3.
