摘要
目的:大鼠心肌缺血后,U50488H激活阿片受体,观察其对心肌缺血大鼠血流动力学及心律失常的影响。方法:建立大鼠心肌缺血再灌注损伤模型,动物随机分为4组,I组:对照组(手术但未实施缺血再灌注及药物干预);Ⅱ组:缺血/再灌注组(I/R);Ⅲ组:I/R+U50488H干预组;Ⅳ组:I/R+U50488H+Nor-BNI共同干预组。试验中监测术前、缺血及再灌注后ECG、心率(HR)、动脉压(ABP)、心输出量(CO)、每搏量(SV)等血流动力学指标;记录各种类型心律失常的发生情况。结果:κ阿片受体激动剂干预组再灌注后ABP、CO和SV降低;心肌缺血前给予阿片受体激动剂,可减少心律失常的发生,应用阿片受体阻断剂Nor BNI后该作用被逆转。结论:阿片受体激动剂U50488H产生了抗大鼠心肌缺血心律失常的作用。
Objective:To monitor the changes of hemodynamics and investigate the effects of U50488H(a opioid receptor)stimulation on myocardial ischemic arrhythmias in rats.Methods:Rats were randomly divided into control,ischemia reperfusion(I/R),U50488H+I/R,and Nor-BNI+U50488H+I/R groups.Electrocardiogram(ECG),heart rate(HR),arterial blood pressure(ABP),cardiac output(CO),stroke volume(SV) and the incidence of ventricular arrhythmia were recorded in rats.Results:ABP,CO and SV were decreased,the chance of myocardial ischemic arrhythmias was reduced with the adminstration of U50488H in rats.The effect of U50488H was abolished by Nor-BNI.Conclusion:U50488H has some direct protective effects during myocardial ischemia and reperfusion in rats and the chance of myocardial ischemic arrhythmias is decreased by U50488H.
