丹参对感染性休克大鼠脑组织保护作用的实验研究 被引量:1

摘要 目的研究丹参对感染性休克大鼠脑组织的保护作用。方法健康雄性Vistar大鼠42只,按随机数字表法分为4个组:正常对照组(n=6)、感染性休克组(n=12)、丹参治疗组(n=12)和丹参联合重组人促红细胞生成素(recombinant human erythropoietin,rhEPO)治疗组(n=12)。除正常对照组外每个组分为2个时间点(6h和12h),各时间点6只。感染性休克组:由大鼠尾静脉注入内毒素的脂多糖(lipopolysaccharide,LPS)5mg.kg-1制备感染性休克模型;正常对照组:注入等量生理盐水代替LPS;丹参治疗组:模型大鼠出现休克症状后立即腹腔注射丹参5mL.kg-1;丹参联合rhEPO治疗组:模型大鼠出现休克症状后立即腹腔注射rhEPO 5 000U.kg-1,半小时后注射丹参5mL.kg-1。术后各组各时间点大鼠分批腹腔麻醉后行眼球摘除术采血及断头取脑组织:血清行血清神经元特异性烯醇化酶(neuron specific enolase,NSE)检测;脑组织匀浆行NO检测;提取脑组织标本进行病理切片,行HE染色和TUNEL染色检查。结果与正常对照组比较,感染性休克组海马部位神经元损伤明显,可见大量TUNEL阳性凋亡细胞,NO、NSE含量显著升高(均P<0.01);丹参治疗组可减轻上述变化,丹参联合rhEPO治疗组更为明显。结论丹参可减轻感染性休克时脑组织损伤及降低海马区神经元细胞的凋亡,其机制可能与抑制NO的过量生成有关。 Objective To study the protective role of salvia on brain of rats with septic shock.Methods Forty-two rats were randomly divided into four groups:control group(n=6),septic shock group(n=12),salvia group(n=12)and combined salvia and rhEPO group(n=12).Rats were injected via the tail vein with lipopolysaccharide(LPS;5 mg·kg-1) to establish the model of septic shock.Whereas the control group was given the same amount of normal saline only.Rats in salvia group were intraperitoneally injected with 5 mL·kg-1salvia immediately when shock signs appeared.Rats in combined salvia and rhEPO group were treated with intraperitoneal injection of 5 000 U·kg-1 rhEPO immediately when shock signs appeared,followed by administration of 5 mL·kg-1 salvia 30 minutes later.At 6 and 12 hours after treatment,eye removal was performed to take blood after anesthesia for neuron specific enolase(NSE) detection and then rats were decapitated to collect brain tissue for NO examination(6 rats per time point in each group).Furthermore,brain sections were stained with HE and were analyzed for apoptosis via TUNEL assay.Results Compared with control group,neurons in hippocampal CA1 region were obviously damaged and the number of TUNEL-positive cells and the levels of NSE and NO were significantly increased by LPS induced septic shock(P0.01).The changes induced by LPS could be significantly reversed by salvia treatment,especially by combined salvia and rhEPO treatment.Conclusion Salvia can alleviate brain damage and reduce hippocampal neuron apoptosis during septic shock,and the mechanism may be involved in the inhibition of excessive NO production.
