摘要
目的研究独活乙醇提取物及不同萃取部位抑制乙酰胆碱酯酶和丁酰胆碱酯酶的活性。方法应用薄层色谱(TLC)-生物自显影技术和微孔高通量筛选的方法。结果在TLC-生物自显影抑酶实验中独活乙醇提取物,石油醚和氯仿萃取部位呈现阳性结果;微孔高通量筛选实验中独活乙醇提取物、石油醚及氯仿萃取部位对乙酰胆碱酯酶(AChE)和丁酰胆碱酯酶(BuChE)均具有抑制作用,浓度为1mg.mL-1时,3个萃取部位对AChE的抑制率分别为(16.88±0.42)%、(11.82±0.55)%、(44.33±3.21)%,对BuChE的抑制率分别为(13.97±1.17)%、(14.64±1.10)%、(18.63±1.24)%。结论独活乙醇提取物、石油醚萃取部位及氯仿萃取部位对2种胆碱酯酶均具有抑制作用,其中氯仿萃取部位对胆碱酯酶抑制率最高,为主要的抑酶活性部位。
Objective To investigate the effect of different extracts of Radix Angelicae Pubescentis on the activities of acetylcholinesterase(AChE) and butyrylcholinesterase(BuChE).Methods TLC-bioautographic assay and microplate high-throughput screening assay were used.Results The ethanol,petroleum ether and chloroform extract from Radix Angelicae Pubescentis had positive result in the TLC-bioautographic assay and microplate high-throughput screening assay.AChE inhibition percentage of these 3 parts at 1 mg·mL-1 was(16.88±0.42)%,(11.82±0.55)%,and(44.33±3.21)%,while BuChE inhibition percentage was(13.97±1.17)%,(14.64±1.10)%,and(18.63±1.24%),respectively.Conclusion The ethanol,petroleum ether and chloroform extract from Radix Angelicae Pubescentis can inhibit both AChE and BuChE.The chloroform extract shows the maximum inhibition.
